You then just need to make a small turn on the focusing wheel rail to move the camera a tiny bit closer for each shot in the series. I wondered about that technique - I have briefly tried using the focus ring but it was so difficult to get a series of closely-enough spaced focussing points.

Depth of Field

A lot of creative focus is waaay overdone IMHO. It doesn't need an awful lot of blur to make your intended object of attention the only one the viewer looks at. The eye is drawn naturally to the sharpest point of the image, especially if it has lead lines or is on a point of thirds. OK theres a problem if you have glaring highlights - but then that's your fault for not composing better ;)

That's a fantastic image Simon. It's a technique I have never tried and you have whetted my appetite. Thanks for that. To clarify for others who may be wondering - the reason for the 1mm/1.5mm advancement of the focus plate is because the depth of field of each shot was only 1.5mm/2mm - so I needed 32 x 'slices' of the flower to be in focus from front to back to give me a composite...

Hi, Interesting sounding project, I believe we have something similar here at the NHM. As you will be reducing the size of the final image then you have the option to crop, the further away you are the greater will be your DoF. I wouldn't rule out a longer FL macro lens, the 90's, 100's are very very usable with DSLR's will allow that little bit extra working distance ( very importatnt...

It may also help to hand tint the parts of the image that are closer to the view with a warm tone and tint the parts that are further away with a cool tone. This lends the illusion of distance. If its subtle enough it will be subliminal.
