This looks fine to me. Your settings for DOF et all seem good too. I would crop the right side red off touching the pink petal for composition, but that is a matter of taste. TG
Thisseems to be OK, so what is happening? This is vry close, so are the problems at longer focal lengths? is itan internal problem with the zoom at certain values?
You need to carry out your tests mirroring the situationswhere the problems seem to arise.

This is really getting good.
I am pretty sure, now, that the problems were to do with quality settings.
Shutter speed and aperture aren't optimum for the light level, but you are now getting good quality for the circumstances, rather than below par.
Now, some more 'real' pictures, maybe?
